中国科研开发出可精准检测与治疗癌症的奈米粒子

新华社6日报导,大陆科研人员成功开发出一种能够实现癌症精准检测与治疗的奈米粒子,可显著降低癌症检测治疗过量使用药物带来的副作用。相关研究成果近日已发表于国际知名学术期刊《先进材料》。
新华社报导,6日从中国科学院精密测量科学与技术创新研究院了解到,该院周欣研究员团队利用肿瘤微环境与正常组织的差异,开发出了一种可智慧识别肿瘤的奈米粒子GQD
NT。这种奈米粒子通过在肿瘤中不断变形,延长了粒子内的药物在肿瘤中的驻留时间、增强了药物在肿瘤中的穿透性,以极低的药物剂量实现了癌症的长时磁共振成像检测与高效治疗。
团队专家指出,药物过量是造成癌症检测与治疗副作用大的主要原因。这是因为现有药物对病灶的靶向不足,难以富集于肿瘤区域,且在病灶部位停留时间短,需要进行大剂量注射以达到预期成像检测与治疗效果。
据悉,GQD NT是一种模组化自组装奈米粒子,可以使用十分简易的步骤将药物分子封装于其中,通过肿瘤微环境促发GQD
NT变形,逐步提高药物在病灶部位的富集浓度。小鼠实验发现,GQD
NT在癌症检测中的造影剂使用量仅为现有临床技术的6%至22%。在注射后4至36小时内,肿瘤部位的造影剂与正常组织对比度高,边界明显,极大延长了磁共振成像时间。
团队基于GQD
NT设计的光动力学治疗方法,单次光动力学治疗后,肿瘤体积下降82%,两次光动力学治疗后,肿瘤被完全消融。实验中,光敏药物的总剂量降至1.76至3.50微摩尔/千克的极低水准,与文献报导相比降低了90%(单次治疗)至95%(两次治疗),且所用的低剂量激光不会造成皮肤损伤,有望克服光动力学治疗中光敏药物过量的问题。
